Evaluating the reliability of the sickling test and peripheral blood film method for screening of sickle cell disease: a study protocol

Background: Mutations in beta-globin are the cause of a widespread condition known as sickle cell disease (SCD). Sickle cells organ damage, hemolysis blood cells, weakness, and sometimes even death. SCD can be effectively managed mortality reduced through early detection. This protocol’s study aims to evaluate sensitivity specificity sickling test, peripheral smear, hemoglobin electrophoresis for screening. Methods: In this study,75 cases will selected. 5ml drawn into dipotassium ethylenediaminetetraacetic acid tube testing SCD. Testing involve: where an equal volume K2EDTA mixed with 2% sodium metabisulfite; Leishman’s stain placed on thin film 2 minutes; Hb electrophoresis, parts distilled water packed combined then mixture centrifuged. Results: We comparing between test method, we conclude depending upon results which is better. Conclusions: Using smear images, quickly diagnosed. A smear-based differential diagnosis may possible but requires special tests such confirm diagnosis. Therefore, help initiate transfusion therapy create better prognosis.
